Abstract:Objective To discuss the effect of “one step method” on constructing GdTetraazacyclododecaneOctreotide
(Gd-DOTA-OCT) small molecular MR imaging probe by adopting Solid Phase Extraction
(SPE)method to isolate it from the mixture. Methods The commercial DOTAOCT
was chelated with
GdCl3 directly, and the mixture was isolated and purified with SPE method, then all the isolated fractions
were underwent ultraviolet spectrophotometric detection, mass spectrometric detection and MR signal
detection respectively. Results GdDOTAOCT
was synthesized with “one step method” and isolated
with SPE method successfully. The actual molecular weight of the probe detected by MALTITOF
was consistent with the theoretic molecular weight, and the GdDOTAOCT
MR probe was
intact after being verified by the ultraviolet spectrophotometric detection and MR signal detection.
Conclusion GdDOTAOCT
small molecular MR probe can be successfully constructed with “one
step method” and can be isolated and purified with SPE method, which lays a foundation for brain tumor
specific imaging.
