Abstract:Objective To investigate the protective effect of self-developed equipment with Nomorthermic machine perfusion (NMP) in different warm ischemic liver. Methods Female Bama pigs were selected as blood and liver donors and were divided into DCD
30 min and DCD 60 min group. NMP equipment was used for perfusion for 12 hours to observe the perfusion parameters, blood gas biochemical indexes and bile-related indexes of the two groups during the perfusion process. Results The hepatic artery flow was
no statistical difference in two groups at different times (P>0.05). The portal vein perfusion pressure in the DCD 30 min group was lower than the DCD 60 min group, and the statistical difference was observed (P<0.05). There was no statistical difference in acidbase
balance, lactic acid, glucose, ALP, γGGT and TBIL between the two groups (P>0.05). The urea nitrogen level was higher in the DCD 60 min group and the transaminase was lower in the DCD 30 min group, and the difference was statistically significant (P>0.05).
Both groups of livers were able to continuously synthesize bile, and there was no statistical difference in pH, bicarbonate and glucose levels (P>0.05). However, the concentration of bile bilirubin and acid was statistically significant (P<0.05). Conclusion At NMP 12
h, the two groups of donor liver with different warm ischemia time were able to conduct aerobic metabolism and continuous bile synthesis, suggesting that the self-developed NMP could effectively protect the donor liver with hot ischemia injury.
