Abstract:Objective To design a standard fluorescent cell counting sample for quality control of automatic fluorescence microscopy
system. Methods Using a custom slide with a black box as the carrier, a certain number of fluorescently labeled human epithelial
cells were fixed in the black box whose surface was treated with polylysine for anti-desorption treatment, then glycerin was added.
Finally, the sample was covered with a cover slip using neutral gum sealant for sealing. The standard samples of various cell
concentrations were prepared and used for accuracy, precision and reproducibility testing of automated fluorescence microscopy
system. Results We prepared three kinds of standard fluorescent cell counting samples, including blank control, positive sample
A, and positive sample B. The cell number confirmed by testing organization was considered as true value. Based on the true value,
the accuracy values of the automatic fluorescence microscopy system for positive sample A and positive sample B were 98.96%
and 97.96%, the precision were 1.92% and 2.00%, and the reproducibility errors were 0.34% and 1.33%, respectively. Conclusion
The designed standard fluorescence cell counting sample has reliable performance and can be used as a quality control tool for the
automatic fluorescence microscopic system.
