Abstract:This paper mainly introduced the components, technical parameters, working principle of
confocal laser scanning microscope Leica SP8 STED 3X equipped in the Instrumental Analysis Center
of Shanghai Jiao Tong University. This microscope, based on the Leica TCS SP8 confocal microscope,
is equipped with super resolution model - stimulated emission depletion, live cell station and two-photon
module; it can be used to carry out nano-level sub-cellular structure and dynamics study, and can also be
used to observe subcellular organelle movement characteristics and changes of living cell, is a ultra-high
resolution confocal microscopy system of various detecting functions. Ultra-high resolution fluorescence
microscopy includes super-resolution technology based on modulation of light spot to reduce point spread
function of the system, as well as super-resolution technology based on single-molecule localization.
Meanwhile, preparation method of the sample, parameter setting principle of the image capture, and
the routine maintenance and management, were also discussed, which has great significance in clinical
practice.
